Embryonic stem cells (ESCs) can differentiate into numerous cell types in vitro, including endothelial cells, hematopoietic cells, cardiomyocytes, and neurons.
During embryonic stem cell culture, maintaining pluripotency—i.e., the undifferentiated state—is critical. ESCs are typically cultured on feeder layers of mitotically inactivated trophoblast or fibroblast cells, or on extracellular matrix-coated surfaces (e.g., fibronectin, laminin, or synthetic basement membrane substitutes).
Serum pre-screening is a mandatory step prior to ESC culture, as it reduces spontaneous differentiation and supports robust proliferation.
